Targeted Screening and Quantification of Pesticide Residuals in Tobaccos by Ultra Fast LC/MS/MS

Although the Method Package developed by Shimadzu includes 167 pesticides from the Positive List, it is often required to update the method by adding more targeted compounds to meet different requirements in different countries. In this study, six additional pesticides, clomazone (240.1>125.1, 240.1>125.1), pendimethalin (282.2>212.1, 282.15>194.1), flumetralin (422.1>143.1, 422.1>107.0), butralin (296.2>240.2, 296.2>222.2), profenofos (370.0>303.0, 373.0>345.1) and chlorpyrifos (350.0>198.0, 350.0>125.0) were added. The updated method was applied to the tobacco samples prepared by QuEChERS method for screening against the 173 pesticides covered by the method. The results showed that three, one and two different pesticides were found in tobacco samples A, B and C, respectively. These positive screening results were significant, because each pesticide was confirmed by two MRM transitions of the method and retention time shift within +/-1.0 min compared to the Method Package. For quantitative determination of the found pesticides, calibration curves were established under the same conditions by running a mixed calibration series in solvent and spiked samples were used as control of the method. For example, the quantification results of sample A were 7.3, 12.3 and 4.8 ng/mL of methomyl, profenofos and chlorpyrifos, respectively. The quantification result of Sample B was 32.2 ng/mL of butralin. This study demonstrates that it is possible to use the Method Package on LCMS-8040 for screening analysis of pesticides (in tobaccos) first and followed by quantification of the found pesticides by setting up calibration for the found compounds. Further study on the reliability of this approach is carried out on more samples.
