Characterization of C-terminal and Disulfide Bond Peptides of Monoclonal Antibody (mAb) on Q-TOF Mass Spectrometer

Sequence analysis of C-terminal and disulfide-bonds linked peptide for monoclonal antibody was evaluated for a representative biosimilars such as bevacizumab by high resolution mass spectrometer, LCMS-9030 Q-TOF. The sample was reduced, alkylated followed by tryptic digestion. Digested peptides are separated by a reversed-phase chromatography and determined by MS/MS fragmentation with the Q-TOF mass spectrometer. All three C-terminal peptides as well as cysteine congaing peptides expected from bevacizumab biosimilar successfully identified on the extract mass chromatogram with accurate masses within 2 ppm.
