Fast and sensitive assay of tobacco specific nitrosamines by UHPLC-MS/MS
Processed tobaccos (i.e. from commercially available cigarettes) were lixiviated with buffer. The resulting liquid sample was then submitted to very selective solid phase extraction on molecularly imprinted polymers. The extract was then evaporated to dryness and reconstituted in buffer. Thanks to the selective extraction almost no matrix effect was measurable. This allowed assaying different sample matrixes with calibration standards made in neat solution. Analysis was performed by reversed-phase UHPLC. Special attention was given to the mobile phase organic solvent and additives to enhance electrospray ionization. Detection was performed with a fast triple quadrupole mass spectrometer. Analysis time was less than 1.5 min. The low limits of quantitation obtained permit to assay a variety of tobacco product and to foresee assay in smoke samples.
