Glycosylation Profile of IgGs Using a Linear Benchtop MALDI-TOFMS and Affinity Purification of Fc

Using MALDI-8020, we tried to profile glycosylation of IgG without a time-consuming release of glycans. To this end, we examined pretreatment in which Fab and Fc are generated by enzymatic cleavage, and the Fc region that undergoes glycosylation is specifically purified with affinity beads. By measuring only the Fc region, we were able to rapidly profile glycans on three types of commercial antibody. Furthermore, we were able to distinguish and classify the three antibody mainly differed in glycans by using the eMSTAT solution.
