Detection of Cysteine/Cystine Using Protein Sequencer

The maturation proteins expressed in a living body function by a thermodynamically stable preferred 3-dimensional conformation depending on the interactions of their constituent amino acid side chains. One of these interactions is disulfide bonding. Determination of the position of the cysteine that comprises the disulfide bond is necessary and indispensable in structural analysis of proteins.
This article introduces an example of an amino acid sequence analysis of a sample containing cysteine and cystine by using the Shimadzu PPSQ-50A protein sequencer systems.
