Shimadzu Releases the Crude2Pure Automated Purification/Powderization System
— World’s First Technology that Automates Fractionation, Purification and Powderization of Target Compounds with High Purity and a Free Base for Improved Productivity —
Shimadzu announces the release of its Crude2Pure (C2P), the world’s first automated purification/powderization system. Incorporating automated purification and powderization technology, using trap concentration*1, this system can purify target compounds from synthetic products consisting of low molecular weight organic compounds and complicated natural substances, and recover them as highly pure powders. It is intended for use in a wide variety of fields, including drug manufacturing, chemistry, foods science, government and academic research, for such applications as the measurement of organic synthetic compounds and natural substances.
By automating purification and powderization, this system significantly reduces operator workload, and provides an environment in which processing is simple even for novices. With this system, Shimadzu provides a solution for preparative, purification and powderization processes when combined with the preparative LC (liquid chromatograph) system, and contributes to improved preparative/purification efficiency.
This system had a reference exhibit at PITTCON 2014, the world’s largest conference and exposition on laboratory science, convened in the United States (March 2 to 6 at the McCormick Place in Chicago, Illinois).
Background to the Development
In the field of organic synthesis, particularly in drug discovery research departments, preparative LC (liquid chromatograph) systems are used for the high-quality purification of synthesized target compounds. However, in the conventional evaporation method the following problems exist when purifying and powderizing the compound from the fractionated liquid obtained: (1) Fractionation and powderization are done by different instruments. (2) If a reversed phase preparative LC system is used, powderization with high moisture content takes long time or longer. (3) Prevention of the powderization originating from the mobile phase used by the preparative LC system and low purity of target compounds.
With its automated purification/powderization technology, the Crude2Pure (hereafter called C2P) system automatically removes components originating from the mobile phase contained in the fractionated liquid. It also enables the powderization of high-purity target compounds in a short time, thus solving the issues associated with the purification/powderization process.
1. World's First: Purification of Target Compounds with High Purity and Free Base*2
|A newly developed trapping column, Shim-pack C2P-H, strongly retains target compounds, and it can easily remove only residual components by rinse washing to purify the sample. Moreover, target compounds can be recovered as free base by rinse washing and counter ion substitution by delivery ammonia water, etc. The free-base compound improves the throughput of biochemical screening and ADME (absorption, distribution, metabolism, and excretion) testing for new drug discovery research.|
2. Powderization of Target Compounds in a Short Time
|This system can powderize target compounds at a high recovery rate by eluting the target compounds retained in the trapping column with high purity in an organic solvent, preventing the splashing powder seen with conventional powderization technology, and volatilizing an organic solvent efficiently. Powderization processes are automated and the sample concentration and powderization process, which requires 8 hours or longer with the conventional evaporation to dryness process, can be shortened to approximately three to four hours.|
3. Powderization of Samples in Open Access Environment
|The Open Solution Crude2Pure Software can set the trap/concentration and powderization operation in three steps and it is easy to check the operating condition of the system. This system is compatible with an open access environment with multiple researchers using the C2P as shared equipment. The sample can be set to C2P and powderized at any time and researchers far from the laboratory can receive notification via e-mail when the powderization ends.|
*1: Trap Concentration
With this technology, the target compound in the fractionated liquid is retained by the newly developed trapping column, which uses a reversed phase resin filling material with a large specific surface area. The target compound is then concentrated at the column tip.

In pharmaceutical development, since a difference in stability of compounds by counter ion species of target compounds, absorption and drug efficacy in the body occurs, it is important to recover target compounds without being influenced by counter ions.
