Precolumn Amino Acid Analysis of Alternative Protein Hydrolysis Samples

Abstract

As the world’s population increases, there is a growing demand for food and an increasing need for sustainable sources of nutrition. Protein sources are particularly important, and the considerable environmental impact of the live-stock industry has prompted a growing interest in alternative proteins. The quality of any protein is influenced by its amino acid composition, hence amino acid analysis is an important tool in determining the quality of protein sources. We measured levels of hydrolyzed amino acids and evaluated the amino acid balance in two of alternative proteins: a soy-based meat and an edible insect food source (cricket powder). Amino acid analysis was performed on an HPLC system with a fluorescence detector by pre-column derivatization with the fluorescent reagent o-phthaladehyde (OPA). The  advantages of this method are short analysis times and a simple analytical process as the derivatization can be performed using the HPLC system autosampler.
