SuperFlo Columns
Radial Flow Columns

Redefining the possibilities in preparative liquid chromatography
SuperFlo® Columns address the fundamental limitation of the axial column design by maintaining low pressure even as column size increases. The unique radial-flow design provides a significantly larger sample inlet area, which results in lower linear velocities, even at high total flow rates. SuperFlo columns are capable of extremely high flow rates and are ideally suited for high-throughput capture applications.

How SuperFlo Columns Work
-
The SuperFlo columns use a radial flow column (RFC) design consisting of two concentric porous cylinders that form the column bed. The space between the inner and outer frits is packed with the chromatographic resin. The eluent or sample enters through the column inlet at the top of the column. The top lid acts as a distribution head, directing the liquid into the space between the outer column body and the outer frit. The liquid is distributed over the entire height of the column and flows horizontally through the packed bed from the outer frit toward the inner core. It then passes into the space between the inner frit and the collector rod and exits through the column outlet at the bottom. The effective bed height is the distance between the inner and outer frits.
The unique radial-flow design of the SuperFlo column provides a significantly larger surface area than traditional axial columns, maintaining optimal linear velocity and effective interaction between the sample and resin even at high flow rates. -

SuperFlo Column Sizes and Materials
-
SuperFlo columns are available in a range of standard sizes and can also be customized to meet specific process requirements. Standard acrylic columns are available in sizes from 50 mL to 1,500 mL with a 3.5 cm bed height. Standard stainless-steel SuperFlo columns range in size from 100 mL to 1,500 mL with a 3.5 cm bed height and are equipped with 10 μm stainless-steel frits. The columns offer excellent chemical compatibility across a pH range of 2 to 12.

Case Study 1: Accelerating Plasma Separation with Radial Flow Columns
SuperFlo columns can significantly reduce processing time while lowering labor and operating costs. The data below were obtained from a plasma fractionation facility and compare the performance of a 20 L SuperFlo column with that of a 16 L axial-flow column. As shown in Table 1, the SuperFlo column achieved more than three times the flow rate, reducing processing time from three shifts to a single shift while maintaining product recovery and purity.

Purification of an Intracellular Bacterial Enzyme
The example below demonstrates the purification of a recombinant protein. During scale-up from a 100 mL axial flow column to a 1,200 mL radial flow column, increasing the flow rate by threefold significantly reduced processing time. The shorter residence time also minimized on-column protein degradation that had occurred during longer processing, resulting in improved product recovery.
