Interactive Optimization with Media Blending Kit

CellTune was used to improve the base culture medium composition for antibody production. To optimize the composition, the following media blending kit explored and optimized the mixture ratio of eight types of supplements in one type of base medium. With culture performance based on antibody production quantity, three rounds of sequential optimization increased the score by about 80 %, which confirmed that the medium composition could be efficiently improved.

- The media blending kit was designed to create diversity in medium composition by allowing users to add any desired amount of supplements to a base medium. Since optimization can be achieved simply by adding supplements to the medium, the kit supports quick and easy workflow improvements.

Designing Experiments with Constraints
The constrained Bayesian optimization function was used to output a design of experiment that maintains a constant total quantity of the base medium and supplements. Using the resulting design of experiment, antibodies were cultured using different blends of the base medium and supplements.

Visualization of Sequential Optimization Results
The visualization function was used to show how much sequential optimization improved culture performance.
The results confirmed that multiple rounds of optimization improved scores by about 80 %, relative to the control condition. The results also showed that average improvements compared to the control condition trended upward as the number of rounds increased.
